- ItemCertificação de métodos microbiológicos de diagnóstico do Laboratório de Controle Qualidade Sanitária do CEMIB/UNICAMP junto ao ICLAS(2016) MOREIRA, Josélia Cristina de Oliveira; DEMOLIN, Daniele Masselli Rodrigues; MINAGAWA, Clarice Yukari; SANTOS, Silvio Rogério Cardoso dos; ANDRADE, Lenira Aparecida Guaraldo de; SILVA, Euma Cunha Ramos; GILIOLI, RovilsonThe international program “ICLAS Animal Quality Network” was developed in 2007 with the aim of improving the quality of animals used in scientific research. Thus, the “Performance Evaluation Program (PEP)” was created to assist diagnostic laboratories in assessing the sensitivity and specificity of laboratory tests. The program has 22 participants worldwide and in South America only the Laboratory of Sanitary Quality Control of CEMIB/UNICAMP is part of this group. Twenty “blind” biological samples were received, consisting of: bacterial cultures isolated from the intestinal tract, fluid and vagina; mouse feces; lung and spleen homogenate; 5 rat sera and 5 mouse sera. For bacteriological diagnosis, enrichment and selective-differential culture media were used. Sera were tested by Indirect Immunofluorescence for 16 viral antigens and tissue samples by PCR and RT-PCR. Of the 20 samples, the laboratory correctly identified 17 of them, which corresponds to 89.4% of diagnostic compatibility. The bacteria identified were: Klebisiella oxytoca, Streptococcus agalactiae, Stenotrophomonas malthophilia, Staphylococcus sciuri and Mycoplasma pulmonis. The sera showed antibodies against: Reovirus-3, mouse minute virus, Mycoplasma pulmonis, murine Hepatitis virus, Sendai virus, Rat coronavirus, Rat Theilovirus and Adenovirus. Of these sera 2 were negative. Molecular analyzes of lung and spleen homogenate showed positivity for murine Pneumocystis and Parvovirus, respectively. This first screening was important to evaluate the adopted methodologies and detect possible problems related to the methods used in the process, ensuring the certification of the quality of the diagnosis offered by the laboratory to the scientific community.
